The emergence of BTK inhibitors and other innovative treatment options has transformed the management of patients with mantle cell lymphoma (MCL) and other B-cell malignancies. Currently, BTK inhibitors and cellular therapy are proven sequential options after progression on upfront regimens (such as R-CHOP and bendamustine/rituximab), but these developments have provoked new challenges, including a need for individualized sequential care in relapsed/refractory (R/R) MCL, therapeutic intolerance and treatment-emergent AEs, and therapy selection after progression on covalent BTK inhibitor regimens.
